• step 1 :

  disinfect the working area
• step 2:

  sterilise the innoculating loop by passing through a bunsen flame
• step 3:

  introduce e-coli onto the plate using the loop
• step 4:

  place filter paper disks onto each different liquid (e.g. dettol, ECP, water)
• step 5

  using a marker pen split the lid of the plate into 4 sections and label each section with the coresponding liquid
• step 6

  sterelise tweezers with the flame
• step 7:

  using tweezers take the filter paper from the liquid and place in correctly labelled sections and replace the lid
• step 8:


  repeat steps 6+7 with the other liquids
• step 9:

  tape the lid shut and place upside down in an incubator at 23'C
• step 10:


  after the bacteria has grown measure the exclusion zone of the circle and calculate the area
