B2 - enzymes/amalyse

  • method:
    1. fill test tube with 50cm of starch solution
    2. place test tube in water bath of 35 degrees
    3. add buffer to change PH to 5
    4. add 10cm of amalyse solution using pipette and start stopwatch
    5. every 30 seconds, use pipette to add drop of solution to well plate - add iodine solution to test for starch
    6. stop when starch solution no longer turns blue-black
    7. repeat 3 times at same PH to calculate mean
    8. repeat for different PH levels
    9. plot graph with PH on x axis and time taken to break down starch on y axis
    • control variables are volume of starch solution and volume of amalyse solution
    • keep time intervals same
