Effect of pH on amylase

  • amylase breaks down starch into simple sugars

    place 1 drop of iodine into each well of a spotting tile

    • fill 1 test tube with 2cm cubed starch solution
    • the second with 2cm cubed amylase
    • the third with 2cm cubed pH buffer

    place the test tubes in a water bath at 30 degrees for 10 minutes so the solutions can reach the right temperatures

    mix the 3 solutions into 1 test tube then place back into water bath and start stopwatch
  • 5
    after 30 seconds transfer a drop of solution to a well of iodine and it will turn blue-black showing starch is present
  • 6
    take a sample every 30 seconds and continue until iodine remains orange which shows starch is no longer present

    repeat with different pH buffers
