Homeganization:
Cells are broken up by homogenization to release organelle content. (Using blender/ hand mixer)
Solution filtered remove large debris
Cell kept in cold (stop enzyme activity), isotonic (prevent osmosis so organelle doesn't burst/shrivel), buffered (pH buffer to prevent organelle damage)
Ultracentrifusion:
Solution spun in centrifuge at high speed.
Dense organelle form pellets at bottom (nucleus) then removed
Supernatant respun at higher speed till all organelle separated (ribosome last)
