How does PCR/invitro cloning work?
Temp of DNA increased to 95C⁰ to break hydrogen bonds, leaving 2 single strands of DNA
Temp decreased to 55C⁰ so primers can attach to each single strand of DNA
DNA polymerase joins free nucleotides by phosphodiester bonds
Temp drops to 72C⁰ so hydrogen bonds can form between the new complimentary strand and the single strand
