  • How does PCR/invitro cloning work?
    • Temp of DNA increased to 95C⁰ to break hydrogen bonds, leaving 2 single strands of DNA
    • Temp decreased to 55C⁰ so primers can attach to each single strand of DNA
    • DNA polymerase joins free nucleotides by phosphodiester bonds
    • Temp drops to 72C⁰ so hydrogen bonds can form between the new complimentary strand and the single strand
  • What are primers?
    • Short single stranded chains of DNA that are complimentary to a longer DNA strand
