• Light-dependant test

  • Isolate chloroplasts in buffer
  • Add DCPIP
  • Electrons from transport chain reduce DCPIP
  • Reduced DCPIP changes colourless to blue
  • Faster colour change faster photosynthesis 
• Light independant test

  • Put leaf in flask with radioactive carbon dioxide
  • Analyse light independant products for radioactive carbon
  • Positive result shows CO2 was used in calvin cycle
