CPAC11: Hill reaction (photosynthesis isolated chloroplast)

• Describe the method

  IV = test tubes

  DV = Time taken for DCPIP to decolourise

  CV = Leaves from same plant species, vol of chilled isolation medium, same time in centrifuge

  Method:

  • In mortar & pestle, grind leaves of plant species
  • Add chilled isolation medium
  • Put solution in centrifuge for set time
  • Extract chloroplast pellet
  • Put supernatant into the test tube (stated in q)
  • Suspend chloroplast pellet in chilled isolation medium
  • Put same vol of sol into test tubes
  • Add DCPIP (expect 1 test tube)
  • Time until DCPIP decolourises
  • Repeat 2 more times + mean time
  • T-test
