CPAC14: Gel electrophoresis

• Devise a method for gel electrophoresis:

  Prior stuff:

  • DNA sample obtained
  • Amplify (PCR)
  • Cut DNA fragments with restriction endonuclease
  • NOW were going to seperate DNA (gel electrophoresis)
  • Compare DNA profile

  Method:

  • Load DNA samples into wells in agarose gel in gel electrophoresis tank
  • Apply electric current across agarose gel for set time (-electrode closest to wells)
  • Fragments seperate according to size
  • Add flourescent dye + shine UV light to visualise the DNA bands
  • Compare DNA bands on DNA profile
  • Look at how similar/different they are
