rp 5: effect of pH on amylase

    1. place one drop of iodine solution into each well of a spotting tile
    2. 1st test tube: 2 cm3 starch solution
    3. 2nd test tube: 2 cm3 amylase solution
    4. 3rd test tube: 2 cm3 pH5 buffer solution - controls pH
    5. place all test tubes in a water bath at 30 degrees & leave for 10 minutes

    1. combine all solutions into 1 test tube & stir w stirring rod
    2. return to water bath & start a stopwatch
    3. after 30s, use stirring rod to transfer 1 drop of solution to each spotting tile
    4. repeat step 3 & continue until the iodine remains orange & record time when reaction has finished
    5. repeat whole experiment using different pH buffers
  • what will happen when the temperature is too high?
    • enzyme will be denatured
    • starch will not be broken down
