  • Step 1: sterilise inoculating loop using a bunsen burner's roaring flame
  • Step 2: Dip disks into antiseptic solutions leaving one out
  • Step 3: Put inoculating loop into bacteria gel
  • Step 4: under the agar plate divide it into the same parts as the disk number with a marker
  • Step 5: open lid slightly next to a bunsen burner (to create a sterile field)
  • Step 6: slowly use the inoculating loop across the plate
  • Step 7: put a disk on one of the divisions
  • Step 8: Close lid and tape it leaving a part tape less (prevents anaerobic respiration)
  • Step 9: store at 25°C for 48 hours upside down (prevent condensation from falling on plate)
  • Step 10: Measure zone of inhibition of each disk
  • Step 11: compare zones of inhibition of each disk
