  • Starch digestion experiment
    1. Place one drop of iodine into each well of a spotting file
    2. Take three test tubes: Test Tube 1=12cm³ starch solution, Test Tube 2=2cm of amylase solution, Test Tube 3=2cm³ of a pH 5 buffer solution
    3. Place all three test tubes in a water bath at 30°C and leave for 10 minutes
    4. Combine all solutions and mix with a stirring rod, return to water bath
    5. Start stopwatch
    6. After 30 seconds, use the stirring rod to add a sample to a well in the spotting file
    7. Continue taking samples every 30 seconds until iodine remains orange, indicating starch is no longer present
    8. Record the time in results
    9. Repeat with pH buffers of different pH
  • Problems only taking samples every 30 seconds- we only have an approximate time. It is not always obvious when iodine stops going blue black
