CP3: Alcohol conc/temp effect on membrane permeability

• Describe the method to investigate effect of temp conc on beetroot

  IV = Temp of water bath 10°C - 50°C

  DV = Light absorbance (colorimeter)

  CV = same amount of water, same amount of beetroot cyclinders, same SA, same type of beetroot, leave test tube in water bath for same amount of time

  Method:

  • Use cork borer to get peices of beetroot
  • Cut into same size - ruler + scalpel
  • Dry with absorbent paper towel
  • Put in test tube
  • Add 10cm3 of H2O (immerse cylinders)
  • Put in water bath
  • Leave for 20 minutes
  • Take tube out
  • Decant liquid only into cuvets
  • Measure light absorbance - colorimeter- blue light filter
  • Repeat 2 more + mean
  • Greater absorbance (au), more pigment betalain, more permeability
