    Primary target: Glucocorticoid receptor
    Action: (Full) agonist
    Secondary target: Mineralocorticoid receptor
    Action: agonist
  • MOA - prednisolone
    Short term effects
    • Decreased vasodilation & permeability of capillaries,
    • Decreased leukocyte migration to sites of inflammation
    Bind to glucocorticoid receptor → mediated changes in gene expression → multiple downstream effects (over hours to days)
    • Inhibit neutrophil apoptosis & demargination
    • Inhibit phospholipase A2decreases formation of arachidonic acid derivativesinhibit NF-kappa B & other inflammatory transcription factors & promote anti-inflammatory genes (e.g. IL-10)
    Low dose → anti-inflammatory effect
    High dose → immunosuppressive
