  • Step 1: Set up water bath at 35°
  • Step 2: put drop of iodine solution in each dimple of spotting tile
  • Step 3:measure 5cm^3 of starch solution and 2cm^3 of buffer solution(record ph) into 2 test tubes
  • Place test tues in water bath and wait for them to reach same temperature as water bath
  • Step 4: add 5cm^3 of amylase solution to one of the test tubes and 2cm^3 of distilled water to other

  • Step: start stopwatch and after 30seconds use 2 pipettes to transfer a few drop of each test tube mixture on spotting tile
  • Step6: obserse if iodine on spotting tile turns blue-black If mixture with amalyse turns iodine blue black the analysis has starch to break down
  • What will control mixture always turn iodine

  • Step 7: repeat steps 4-6 every 30 seconds until mixture with amylase stops turning iodine blue-black
  • Step 8: record time it took for iodine to stop turning blue-black in results table
  • Step9: repeat method for remaining buffer solutions
  • The shorter the time the faster amylase has worked to break down starch
