  • Notch is a receptor (transmembrane protein) that is made and trafficked to the membrane
    • its ligand is Delta (transmembrane protein on another cell)
  • Notch starts at the golgi lumen -> cleaved at site 1 -> overlap at the cut site to trigger plasma membrane transport to be an active receptor where it interacts with delta ligand signal on another cell to cause a conformation change on Notch which causes 2 things => proteolytic cleavage of extracellular domain and endocytosis of the remaining receptor which undergoes conformation change for proteolysis of Notch tail -> the cleaved Notch tail fragment enters the nucleus -> joins protein complex with Rbpsuh -> binds to Notch responsive elements to act as co- activator for Notch target genes
