  • Effect of pH on amylase. what do we need
    -Iodine solution
    -starch solution
    -buffer solution
    -amylase solution
    -test tubes
    -pipettes
    -water in a beaker
    -spotting tile
  • Steps
1.Pour equal amount of starch, amylase and buffer solution to each test tube.
2.Place the test tube in a water bath.
3.After a while, add all of the solutions together and let it sit in the water.
4.Using a pipette, add a drop of the solution into the spotting tile which has iodine solution.
5.Add drops to next holes in the spotting tile every 30s, measure with a timer.
6.When the solution in the spotting tile doesnt turn black/blue but stays orange it means starch is no longer present and that amylase broke it down?
  • Why is water bath needed
To make the solutions the right temperature
  • Whats buffer solution

    A solution that controls pH
  • How many of each solution (amylase, starch, buffer) do we need?
    2cm cubed
