  • Cold- Reduce enzyme activity which may break down organelles
  • Isotonic- Prevent organelles from bursting
  • Buffered- Maintain constant pH
• Ultracentrifugation

  1. Homogenising breaks up tissue and releases organelles
  2. Homogenate is filtered to remove large debris which sink to the bottom of the test tube
  3. Centrifugation
