  • Microscopic Examination Procedure for Plants
    1. Calibrate eyepiece graticule with stage micrometer.
    2. Cut thin transverse sections of plant stem on a white tile.
    3. Place section on slide, prevent dye spread with wax crayon, add Phloroglucinol (detects lignin), and cover slip.
    4. Set microscope to lowest magnification, adjust coarse knob.
    5. Use fine knob for clear focus, switch to higher magnification if needed.
    6. Observe and draw stem.
    7. Measure stem diameter and vascular bundle using calibrated eyepiece graticule.
